Registering a Protein
Proteins have to be registered and approved before you can use them when registering samples or building a shipment.
Registration
Register your protein through LS-CAT's protein/proposal registration process. Once approved, it becomes available in ISPyB/Synchweb for shipping and data collection.
Finding it in ISPyB/Synchweb
Select Proteins from the proposal dropdown menu to see your registered proteins. Approved proteins show a green label; use the filter box to search by name.
Names must match exactly
Protein names in your registration and in ISPyB/Synchweb have to match. If they don't, your samples won't be available when you go to build a new container.
Add a sequence or a PDB file
Supplying extra information about your protein unlocks automated analysis after data collection:
- Sequence — enables automated molecular-replacement (MrBUMP) and lets a predicted structure be generated (e.g. via AlphaFold, where enabled) ahead of your visit.
- PDB file — enables automated refinement (DIMPLE) once data has been collected.
Cloning a protein
If you're working with a variant of a protein you've already registered — a selenomethionine derivative or a point mutant, for example — select the approved protein, click Clone Protein, edit the details, and click Add Protein to save the new entry.
